بررسی تأثیر تنش شدت‌نور در ذخیره بتاکاروتن توسط کشت‌‌خالص و مختلط میکروجلبک

چکیده

بتاکاروتن یک رنگدانه قرمز ـ نارنجی است که در گیاهان و میوه ها یافت می‌شود. یکی از بهترین منابع تولید بتاکاروتن میکروجلبک‌ها می‌باشند. تاکنون تولید بتاکاروتن در نمونه‌های خالص بررسی شده است. به دلیل مشکل ­های عملی کشت‌های خالص، دراین مطالعه تنش شدت نور بر نمونه‌های کشت مختلط دریاچه خزر و میکروجلبک خالص دونالیلا سالینا اعمال و چگونگی رشد، میزان کلروفیل تولید شده و تجمع بتاکاروتن در گونه‌های مورد آزمایش مورد بررسی قرار گرفت. در این پژوهش ، شرایط عملیاتی آزمایش برای هر دو نمونه یکسان با 5/7  pH و دور هم­زدن rpm 160 بود. تنش نوری در شرایط عملیاتی یکسان و دمای  oC 28 – 26 در روز ششم اعمال شد. با اعمال این تنش مقدار بتاکاروتن در میکروجلبک دونالیلا سالینا از مقدار اولیه 9/5 به مقدار پایانی mol Beta-Carotene/g Protein 5/19 و در میکروجلبک دریاچه خزر از مقدار اولیه 3/7 به مقدار پایانی mol Beta-Carotene/g Protein 7/22 رسید. غلظت پروتئین در آغاز و پایان فرایند کشت‌های خالص و مختلط به ترتیب از 3/467 به  mg/L 9/1239 و از 2/495 به mg/L 3/1131 رسید. نتیجه­ های به دست آمده از این مطالعه، پتانسیل بالای میکروجلبک کشت مختلط دریاچه خزر به عنوان جایگزین مناسب میکروجلبک خالص دونالیلا را نشان می‌دهد. درنتیجه تولید بتاکاروتن با استفاده از میکروجلبک‌های مختلط دریاچه‌ی خزر که نیازی به استریلیزاسیون ندارد، اقتصادی‌تر از نمونه خالص بوده و به دلیل هزینه‌های کم­تر، امکان صنعتی شدن آن افزایش می‌یابد
