EETs have a wide range of physiological effects. Among them, vasodilatory actions in vascular conduit, renal afferent arterioles, and coronary vessels are more considerable and lead to the regulation of blood pressure and myocardial perfusion. In addition, EETs modulate adhesion molecule expression, platelet aggregation, vascular smooth muscle cell migration, and thrombolytic properties, which could be involved in a protective mechanism against athero 5 sclerosis. Therefore, sEH inhibition that leads to the accumulation of active EETs, provides a novel approach to the treatment of hypertension and atherosclerosis. Since the most reported potent sEH inhibitors have limited pharmacokinetic profile, they aren’t useful for clinical application. The effort to achieve to sEH inhibitors with proper potency and enhanced pharmacokinetic properties is still continuous. In this study, new urea-based compounds with oxadiazole ring against sEH enzyme were developed. The designed compounds showed a high affinity to the active site of the sEH enzyme and were synthesized in good yield and characterized by IR, Mass, and 1HNMR. Some novel compounds had comparable in vitro sEH inhibitory activity to 12-(3-Adamantan-1-yl-ureido)-dodecanoicacid (AUDA), a potent urea-based sEH inhibitor.}, keywords = {Inhibitor,antihypertensive agents,soluble epoxide hydrolase,urea,oxadiazole}, title_fa = {طراحی، سنتز و ارزیابی زیستی مهارکننده‌های نوین اوره ای آنزیم اپوکساید هیدرولاز محلول}, abstract_fa = {آنزیم اپوکساید هیدرولاز محلول (sEH)، اپوکسی ایکوزاترئونیک اسید (EETs) ایجاد شده توسط اپوکسیژناز از آراشیدونیک اسید را تبدیل به شکل دیول می‌کند. اپوکسی ایکوزاترئونیک اسید دارای بازه بالای فعالیت فیزیولوژیکی می باشد که در این میان، گرفتگی رگ ها در مجرای عروقی، شریان اورنده کلیوی و شریان های کرونر از اهمیت ویژه ای برخور دار است که منجر به تنظیم فشار خون و پرفیوژن میوکارد می شود. همچنین، اپوکسی ایکوزاترئونیک اسید قادر به تنظیم بیان ملکول های چسبندگی، تجمع پلاک تها، مهاجرت سلول های عروقی عضلههای صاف و ویژگی های ترومبولیتیک می‌باشد که در گیر در مکانیسم های محافظت شریانی بر علیه آترواسکلروزیس میباشد. بنابراین، مهار آنزیم اپوکساید هیدرولاز محلول منجر به انباشتگی این سوبسترا در بدن می‌شود که می تواند رویکرد نوینی در درمان عارضه فشار خون و آترواسکلروزیس باشد. از آن جا که بیشتر مهارکننده های گزارش شده تا به حال، ویژگیهای فارماکوکنتیکی نامناسبی داشتهاند و برای کاربرد بالینی دلخواه نمی‌باشند. تلاش برای معرفی مهارکنندهای نوین این آنزیم با قدرت مناسب و ویژگی های فارماکوکینتیک بهبود یافته همچنان ادامه دارد. در این پژوهش،ترکیب های جدید اکسادیازولی دارای گروه اوره ای به منظور مهار آنزیم sEHمعرفی شده اند. ترکیب های طراحی شده در مطالعه های داکینگ تمایل بالایی به جایگاه فعال آنزیم نشان داده و با بازده خوبی سنتز و توسط IR و Massو 1HNMR تأیید ساختاری شد. برخی از ترکیب های سنتز شده در ارزیابی زیستی، فعالیت مهاری قابل قیاسی با 12- (3-آدامانتان – 1 – ایل - یوریدو) - دودکانوئیک اسید (AUDA)، مهارکننده قوی اوره‌ای اپوکساید هیدرولاز محلول، داشتند.}, keywords_fa = {مهارکننده,عامل های ضد فشار خون,اپوکساید هیدرولاز محلول,اوره ا,اکسادیازولی}, url = {https://www.nsmsi.ir/article_30923.html}, eprint = {https://www.nsmsi.ir/article_30923_4e9d649a1cd0012650da0498b884417c.pdf} }
