نانوصفحات گرافن اکسید به عنوان حامل های دارویی بورتزومیب در یک سیستم دارورسانی هدفمند: رویکردی جدید برای درمان سرطان

چکیده
این مطالعه برای اولین ­بار بر روی برهمکنش داروی ضد سرطان بورتزومیب و نانو گرافن اکسید توسط محاسبات مکانیک کوانتومی با استفاده از نظریۀ تابعیت چگالی و محاسبات  NBO  با سطح تئوری  B3LYP/6-311+G**  و مکانیک مولکولی با روش مونت کارلو در میدان­ های نیروی AMBER, OPLS, CHARMM (Bio), MM+ در گستره­ای از دما ( 316-298کلوین) و حلال آب انجام شده است. محاسبات نشان داده است که در نانو گرافن اکسیدی، گروه­ های هیدروکسیل و اپوکسی با گروه عاملی N-H  مولکول بورتزومیب پیوند هیدروژنی تشکیل می ­دهند و این در حالی است که گروه کربوکسیل در لبه­ های صفحۀ نانو گرافن اکسیدی بار سطحی منفی ایجاد می ­کند. از این­رو، نانو گرافن اکسیدی به خوبی در آب و سایر حلال­ های قطبی پراکنده می ­شود و همین ساختار دوگانه­ دوست در مواجه با داروی بورتزومیب، آن را به عنوان یک حامل مناسب معرفی می­ نماید، این مهم طبق شبیه­ سازی­های داکینگ مولکولی نیز با برقراری پیوندهای هیدروژنی دقیق و پیوندهای پای آب­گریز این سیستم دارویی ملاحظه می­گردد، که به سبب پیوند­های هیدروژنی، دارای یک پایداری درون مولکولی بورتزومیب باگرافن اکسید و به واسطۀ پیوند­های آب­گریز، تثبیت و پایداری برون مولکولی این سیستم دارویی با محیط اطراف برقرار می­ باشد. مطابق با نتایج NBO  که مبنی بر  برهمکنش π-π گرافن اکسید و بورتزومیب است، برهمکنش های واندروالسی، یونی و هیدروژنی به دلیل حضور تعداد زیادی گروه­ های اکسیژن­دار اتفاق می­ افتد که این امر تاثیر شگرفی در تثبیت داروی ضد سرطان بورتزومیب خواهد داشت. همچنین ارزیابی­ های پتانسیل الکتروستاتیک مولکولی  و بارهای مولیکنی با بررسی برهمکنش­ های الکتروستاتیک این سیستم دارویی از لحاظ اصل الکترونگاتیویته، مطلوبیت و پایداری BTZ-GO را نشان داده و تثبیتی بر نتایج NBO  هستند. محاسبات مونت کارلو نیز نشان می ­دهد که نتایج مستخرج از میدان نیرو Amber  بر نتایج مکانیک کوانتومی منطبق است.
