ارزیابی ویژگی ضدمیکروبی نانوکامپوزیت نقره/ آهن اکسید تولید شده با نشاسته

چکیده

سامانه دوجزئی نقره و آهن اکسید به دلیل تلفیق ویژگی­ های این نانوذره­ های با یک­دیگر مانند ویژگی مغناطیسی، نوری و ضدمیکروبی در حوزه­ های گوناگون بویژه پزشکی توجه زیادی را به خود جلب کرده است. در این پژوهش، نانوکامپوزیت نقره/آهن اکسید (Ag/Fe3O4) با استفاده از نشاسته به عنوان عامل پایدارکننده و متصل کننده این دو ذره به یک­دیگر سنتز شد و فعالیت ضدباکتریایی و ضد فیلم زیستی آن علیه باکتری­ های اشریشیاکلی، سودوموناس آئروژینوزا و استافیلوکوکوس اورئوس مورد بررسی و مقایسه با نانوذره­ های نقره قرار گرفت. نانوذره­ های نقره موجود در نانوکامپوزیت به­ صورت جداجدا با اندازه nm50-15 به­ واسطه گروه ­های عاملی نشاسته به توده­ ای از نانوذره­ های Fe3O4 با اندازه nm10-2 متصل شدند. MIC و MBEC نانوکامپوزیت بسته به گونه باکتریایی و شرایط تولید به ترتیب در بازه 14-2 و 30-20 میلی­ گرم بر لیتر به ­دست آمد. نصف مقدار MIC نانوکامپوزیت باعث افزایش فاز تاخیری منحنی رشد اشریشیاکلی به مدت 8 ساعت و استافیلوکوکوس اورئوس به مدت 5 ساعت شد. نانوذره ­های آهن اکسید به تنهایی تأثیر محسوسی بر منحنی رشد باکتری­ های گرم مثبت و منفی مورد بررسی نداشتند، اما در نانوکامپوزیت باعث ایجاد اثر هم ­افزایی بر فعالیت ضدباکتریایی نانوذره­ های نقره علیه باکتری گرم منفی شدند. نانوکامپوزیت Ag/Fe3O4  در غلظت نصف MBEC نیز برخلاف نانوذره­ های نقره باعث ثابت نگه داشتن جرم فیلم زیستی شد. بررسی زمان مرگ باکتری­ ها نشان داد که نانوذره ­های نقره موجود در نانوکامپوزیت به دلیل درگیری با نشاسته و نانوذره­ های آهن اکسید و متعاقبا رهایش کندتر نقره در مقایسه با نانوذره ­های نقره مجزا با سرعت کمتری باعث مرگ باکتری­ های گرم مثبت و منفی می ­شود.
